Mechanism of action:

Doramectin is a class of macrocyclic lactone anthelmintic that enhances the release of GABA at presynaptic neurons. GABA acts as an inhibitory neurotransmitter and blocks the post-synaptic stimulation of the adjacent neuron in nematodes or the muscle fibers in arthropods. By stimulating the release of GABA and or producing a slow and irreversible increase in permeability of glutamate-gated chloride channels (GluCl) to chloride ions and hyperpolarization of nerve cells, doramectin causes flaccid paralysis of the parasite and eventual death. Tapeworms (Cestoda) and flatworms (Trematoda) (such as Fasciola species) do not use GABA as a chemical mediator in peripheral nerves, so this drug has no effect on these parasites.

Dosage & Administration:

A single treatment of 1 ml (5 mg)/10 kg B.W., applied topically along the mid-line of the back in a narrow strip between the withers and tail head.

Withdrawal time:

Meat: 35 days

Do not use in cattle that produce milk for human consumption.

Doramectin is very toxic to aquatic organisms and may accumulate in sediments. The risk to aquatic ecosystem will be reduced by keeping treated cattle away from water bodies for 2 to 5 weeks after treatment.
